Large protein - Advice?

Hi, I have a large protein (over 3000 residues) and I am looking to see how a mutation (in the 336 aa) will impact the structure. Because I-Tasser can only run 1500 residues I planned to use ThreaDom to find the domains of the protein and run I-Tasser on the domain the muation is in. However the protein came back as one domain. Does anyone have any advice on what I could do? I am under the impression that running I-Tasser for the first 1500 residues is not accurate but I am not sure how else to do it!?

If anyone has any advice it would be much appreciated! Thanks!

Dear user,

you can try https://zhanggroup.org/FUpred to predict the domain and use https://zhanggroup.org/D-I-TASSER to predict the domain level models.

Also, you can try https://zhanggroup.org/I-TASSER-MTD/
